Pyrimethamine and sulfadoxine may decrease body folate levels if taken with the following medications: Cyclosporin and Trimethoprim. Sulfadoxine when given together with the antiepileptic drug phenytoin may increase the level of phenytoin in the blood and may also enhance the effects of the drug in lowering the levels of folic acid in the blood.Sulfadoxine may increase the risk of developing a disturbed heart rhythm when given with amiodarone. These medicines should not be used together. Sulfadoxine may increase the blood thinning effects of nicoumalone and warfarin. Sulfadoxine when given together with methotrexate may enhance the antifolate effect of the two drugs and may lead to toxic side effects. How does Fansidar tablets work?

Fansidar tablets contain two active ingredients, pyrimethamine and sulfadoxine. Pyrimethamine is an antimalarial medicine and sulfadoxine is a type of antibiotic called a sulphonamide. Pyrimethamine and sulfadoxine work by blocking the production of a chemical called folinic acid inside the parasite. Folinic acid is essential in the production of new genetic material (DNA). DNA is necessary for the parasite to reproduce, therefore by blocking it's formation this prevents the malarial parasites from increasing in number. This combination is commonly used in conjunction with quinine in the treatment of malaria. It is not recommended for the prevention of malaria.
